Overview

Cognitive performance is not a single trait but an interaction of focus, working memory, processing speed, mood regulation, and long-term consolidation. A protocol attempting to improve "cognition" should therefore target multiple systems rather than chasing a single neurotransmitter.

This protocol differs from the introductory Nootropic Stack Protocol in that it layers three peptide tools with distinct mechanisms: Semax for focus and neurotrophic support, Selank for anxiolysis and cognitive buffering under stress, and Dihexa as a potent synaptogenic compound used more conservatively due to its strength and limited long-term safety data.

The target user is someone with a high cognitive-demand schedule (knowledge work, graduate study, intensive creative work) who has already addressed sleep, exercise, and diet as the foundational performance inputs.

Semax

Semax is used as the daily workhorse for focused cognitive work. It has a mild, non-jittery focus effect and upregulates BDNF and NGF in preclinical research. Morning dosing aligns with peak cognitive demand.

Selank

Selank complements Semax by taking the edge off the sympathetic tone that high-stakes cognitive work can produce. It is particularly useful for presentation, interview, and deadline scenarios where anxiety is performance-limiting.

Dihexa

Dihexa is potent and less validated than Semax/Selank. It is included here with careful framing: low-dose, intermittent use, during high-demand phases only, rather than daily chronic administration.

Protocol Structure

Phase 1 — Foundation (Weeks 1–2)

  • Sleep 7.5–9 hours, consistent schedule, dark cool room — non-negotiable prerequisite
  • Zone 2 cardio 3x/week, resistance training 2–3x/week
  • Morning light exposure within 30 minutes of waking
  • Alpha-GPC 300 mg with breakfast
  • Creatine 5 g daily
  • Omega-3 2 g/day with food

Phase 2 — Daily Peptide Layer (Weeks 3–8)

Morning (upon waking or with coffee):

  • Semax 200–400 mcg intranasal (2–3 sprays per nostril depending on concentration)

Midday (pre-deep-work block):

  • Semax 200 mcg top-up, optional
  • Or Selank 200–400 mcg if work requires calm precision (writing, editing, negotiations)

Afternoon (3–5 PM):

  • Selank 200 mcg intranasal if residual anxiety or to facilitate later wind-down

Avoid late-day Semax to prevent sleep interference.

Phase 3 — Advanced Layer (Selective, 2–4 week blocks)

Run during discrete high-demand phases (thesis defense, launch week, exam period). Do not run continuously.

  • Dihexa 8–10 mg/day oral, with a fat-containing meal, for 2–4 weeks
  • Continue Semax / Selank as in Phase 2
  • Follow each Dihexa block with a minimum 4 weeks off

Cycling and Duration

  • Semax / Selank: 8 weeks on, 2–4 weeks off
  • Dihexa: 2–4 weeks on, ≥4 weeks off, no more than 2 cycles per year
  • Foundational supplements (creatine, omega-3, choline) run continuously

Important Considerations

  • Peptides do not substitute for sleep, training, and dietary quality. Sleep-deprived peptide users get a fraction of the benefit.
  • Dihexa is potent and less studied in humans; long-term safety data is sparse. Conservative dosing and duration are strongly recommended.
  • Semax can cause mild headaches, irritability, or sleep disruption at higher doses or late dosing.
  • Selank generally lacks stimulant side effects but can occasionally produce mild fatigue when overused.
  • Individuals with bipolar spectrum disorders, psychotic disorders, or uncontrolled anxiety disorders should not self-manage with neuropeptides; consult a psychiatrist.
  • Intranasal formulations must be stored correctly (refrigerated) and dosed consistently; concentration drift between batches is common in research-grade products.
  • Cognitive metrics are subjective; track performance with a simple workload journal rather than trusting felt effects.
